چکیده

Background: Fitness loss and compensation in fusidic acid resistant EF-G mutants of Staphylococcus aureus. Results: Slower translocation and ribosomerecycling due to restricted conformational change, plus increased tRNA drop-off cause fitness loss in F88L; these are recovered in F88L+M16I leading to fitness compensation. Conclusion: Conformational dynamics of EF-G is crucial for its function. Significance: Provides insight into how antibiotic resistant mutations affect in vivo fitness.

Compensatory adaptation to the loss of biological fitness associated with acquisition of fusidic acid resistance in Staphylococcus aureus.

Recent studies have shown that individual amino acid exchanges within elongation factor G (EF-G) cause fusidic acid resistance in Staphylococcus aureus. The data from the present study illustrate that the fusidic acid resistance-mediating amino acid substitutions P406L and H457Y are associated with a marked impairment of the biological fitness of S. aureus. In particular, strains producing EF-G...

Antimicrobial activity and mechanisms of resistance to cephalosporin P1, an antibiotic related to fusidic acid.

The antimicrobial properties of cephalosporin P1, an antibiotic structurally related to fusidic acid, were examined. Cephalosporin P1 exhibited potent activity against methicillin-sensitive Staphylococcus aureus, methicillin-resistant S. aureus and vancomycin-intermediate S. aureus. Mutants of S. aureus resistant to cephalosporin P1 arose with a frequency of 1.6 x 10(-6) for selections at 4 x M...

Anti-staphylococcal activity of indolmycin, a potential topical agent for control of staphylococcal infections.

OBJECTIVES We sought to investigate the anti-staphylococcal activity of indolmycin, with particular emphasis on comparing its activity with fusidic acid and mupirocin. METHODS Established procedures were used to examine the activity of indolmycin against a range of clinical isolates, including strains resistant to fusidic acid and mupirocin. Indolmycin-resistant mutants were recovered and cha...

Fusidic acid resistance in Staphylococcus aureus nasal carriage strains in nine European countries.

AIM To evaluate fusidic acid resistance pheno- and geno-typically in nasal carriage Staphylococcus aureus isolated from general practice patients in nine European countries. MATERIALS & METHODS Phenotypic fusidic acid resistance was determined by disc diffusion and MIC values, and genotypically by a PCR detecting fusA-E genes. The main analysis was performed on methicillin-susceptible S. aure...

Detection of fusidic acid resistance determinants among Staphylococcus aureus isolates causing skin and soft tissue infections from a tertiary care centre in Chennai, South India

Background Fusidic acid (FA)an inhibitor of protein synthesis has been used for treating superficial and some systemic infections caused by Staphylococcus aureus. Fusidic acid resistance in S. aureus has been reported throughout the world with prevalence ranging from 0.5% to 50% and is due to i) point mutation in bacterial fusA or fusE gene and ii) by acquired FA resistance determinants fusB, C...
